Flotillin-1 Antibody (F-3) is a mouse monoclonal IgG1 antibody that detects Flotillin-1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Flotillin-1 plays a crucial role in organizing and functioning of lipid rafts, which are specialized membrane microdomains rich in sphingolipids and cholesterol, essential for various cellular processes including signal transduction and membrane trafficking. Flotillin-1 is primarily localized in the plasma membrane, where Flotillin-1 contributes to forming lipid rafts that facilitate clustering of signaling molecules, thereby enhancing cellular communication efficiency. Flotillin-1 is part of the band 7.2/stomatin protein family and is characterized by a hydrophobic N-terminal region that forms a transmembrane domain, allowing anchoring within the lipid bilayer. Flotillin-1 expression is particularly notable in A498 kidney cells, muscle cell lines, and fibroblasts, and forms a ternary complex with CAP and Cbl, which is vital for directing localization of signaling complexes to lipid raft subdomains. Additionally, Flotillin-1 interaction with other proteins, such as ER-X, is significant for rapid activation of MAP kinase cascade by estrogen, highlighting importance in cellular signaling pathways. Flotillin-1 expression levels are independently regulated and have been correlated with progression of Alzheimer′s pathology, making Flotillin-1 a valuable target for research in neurodegenerative diseases.
