Fibronectin Antibody (IST-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects Fibronectin in mouse, rat, human, porcine, and canine samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-Fibronectin antibody (IST-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Fibronectin is a crucial extracellular matrix glycoprotein that plays a significant role in cell adhesion, motility, and wound healing, making Fibronectin vital for tissue repair and regeneration. This high molecular weight heterodimeric protein is predominantly located on cell surfaces, in extracellular fluids, and in plasma, where Fibronectin facilitates interactions between cells and the extracellular matrix. Alternative splicing and glycosylation of Fibronectin lead to the generation of various isoforms, some of which are associated with specific tissues or pathological conditions, such as malignancies. The presence of glycosylated Fibronectin and Fibronectin splice variants has been linked to the formation of the myofibroblast phenotype in Dupuytren′s disease, highlighting the importance of understanding post-translational modifications in disease contexts.

For IST-9 there is a picture of kidney tissue stained for IHC using IST-9 antibody. Can you please give me the reference for the image as i am looking for the IHC protocol used to produce this image i.e. processing and antigen optimisation process used.

Does this antibody work for IF of formalin-fixed paraffin-embedded tissue?

Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue?

Thank you for your question. For IHC, we recommend using a directly conjugated mouse monoclonal primary antibody. We recommend using SC-8422 for Fibronectin antibody conjugates, including AC, HRP, FITC, PE, Alexa Fluor® 488 and Alexa Fluor® 647. Rated 2 out of 5 by from no bandI used this antibody at 1:200 - 1:1000 as 1st antibody with 2nd mouse antibody at 1:1000 - 1:5000. Both 1st and 2nd antibody I dilute with 5% skim milk but still doesn't work. even on semi-dry or wet transfer also showed no band. Any kind suggestion please?

Rated 2 out of 5 by from Strong background in IF on rat frozen liverRat liver frozen sections were subjected to immunofluorescence using Fibronectin (IST-9) antibody. Antigen retrieval methods were optimized between without antigen retrieval, heat-induced citrate buffer, and trypsin-induced antigen retrieval. Some signals could be observed in portal tracts, but background was strong.

Rated 3 out of 5 by from Staining of distal colon for fibronectin EDAI've had the chance to test the antibody for the staining of fibronectin EDA in the distal colon of mice that were developing fibrosis in the colon. Unfortunately, we did not see staining of the paraffin embedded slides, which also means that there is no background staining and the antibody is very specific. We are currently still trying to change some aspects of the staining protocol to check if we can get better staining.
