FGF6 encodes fibroblast growth factor 6 (FGF-6), a secreted signaling ligand that primarily engages FGFRs to activate downstream MAPK/ERK, PI3K/AKT, and PLCγ pathways. It contributes to regulation of cell proliferation, survival, and differentiation, with prominent roles in skeletal muscle development, regeneration, and tissue remodeling. Altered FGF6 signaling can perturb growth factor–driven transcriptional programs and extracellular matrix–associated processes, making it relevant to studies of dysregulated proliferative signaling and lineage specification. In human cell systems, FGF-6 provides a tractable node for interrogating growth factor crosstalk and context-dependent FGFR pathway output.
FGF-6 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the FGF6 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the FGF6 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the FGF6 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish FGF-6 protein expression.
This CRISPR knockout system enables efficient generation of FGF6-deficient cell models for investigation of FGF-6 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
