FAM127B CRISPR/Cas9 KO Plasmid (h) | sc-406247 | 20 µg | $397.00 |
FAM127B (family with sequence similarity 127 member B) encodes a poorly characterized human protein with limited functional annotation, making it of interest for systematic genetic interrogation. Available evidence suggests roles in cellular homeostasis and gene regulatory processes, with potential connections to signaling and transcriptional programs that influence proliferation and stress responses. As with other understudied FAM family members, altered expression or copy number has been reported in subsets of human disease datasets, supporting investigation in cancer and other complex phenotypes. Dissecting FAM127B function can help clarify its contribution to pathway wiring, cell-state maintenance, and context-dependent vulnerabilities.
FAM127B CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the FAM127B gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the FAM127B together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the FAM127B open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish FAM127B protein expression.
This CRISPR knockout system enables efficient generation of FAM127B-deficient cell models for investigation of FAM127B signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
