F4/80 Antibody (BM8) is a rat monoclonal IgG2a antibody that detects the EMR1 protein of human origin, F4/80 protein of mouse origin, and the corresponding rat homolog by western blotting (WB), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). F4/80 (BM8) antibody is available in a non-conjugated form, making F4/80 (BM8) monoclonal antibody versatile for various experimental setups. F4/80 protein, also known as EGF-like molecule containing mucin-like hormone receptor 1, is a member of the epidermal growth factor (EGF)-TM7 family, which includes several class B G-protein coupled receptors predominantly expressed on immune cells. Notably, F4/80 is exclusively found on the surface of macrophages, serving as a critical marker for mature macrophage populations in tissues such as Kupffer cells in the liver, splenic red pulp macrophages, brain microglia, gut lamina propria, and Langerhans cells in the skin. F4/80 protein undergoes significant post-translational modifications, including extensive N-linked glycosylation and some O-linked glycosylation, which are essential for stability, localization, and function. While the precise role of F4/80/EMR1 remains to be fully elucidated, F4/80 plays a vital part in macrophage adhesion, migration, and potentially in G-protein coupled signaling pathways, highlighting F4/80′s importance in immune responses and tissue homeostasis.

I will use it for formalin-fixed mouse sections. Does citrate unmasking buffer work besides EDTA?

How many tests could i get out of 100ug with a 1:250 dillution

Thank you for your question. This antibody is provided at 100µg in 1ml. In dilutions, you multiply the dilution factors for each step. The dilution factor or the dilution is the initial volume divided by the final volume. (DF=Vi/Vf) For example, if you add a 1 mL sample to 9 mL of diluent to get 10 mL of solution, DF=Vi/Vf = 1mL / 10mL= 1/ 10. This is a 1:10 dilution.

Each vial contains 100 µg of F4/80 (BM8) mouse monoclonal IgG2a in 1.0 ml of PBS with < 0.1% sodium azide and 0.1% gelatin.
