Estrogen Receptor alpha Antibody (H222) is a rat monoclonal IgG1 raised against human Estrogen Receptor alpha (ERα), and detects ERα of mouse, rat, and human origin by western blotting (WB). Anti-ERα antibody (H222) is available in non-conjugated and multiple conjugated forms including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates. ERα protein is a ligand-activated transcription factor belonging to the steroid/thyroid hormone receptor superfamily, playing a critical role in regulating the normal function of reproductive tissues. Post-translational modifications (PTMs) of ERα, such as phosphorylation, ubiquitination, sumoylation, acetylation, and methylation, are crucial for modulating activity, stability, cellular localization, and interactions with co-regulatory proteins. For instance, phosphorylation at specific serine residues can enhance or repress ERα′s transcriptional activity, influencing gene expression involved in cell proliferation and differentiation. Ubiquitination targets ERα for proteasomal degradation, thereby controlling receptor levels within the cell and affecting hormonal responsiveness. These PTMs provide dynamic regulation of ERα functions in response to hormonal and environmental signals. Dysregulation of these modifications has been implicated in the development and progression of hormone-dependent cancers, such as breast cancer. ERα monoclonal antibody (H222) is an invaluable tool for researchers studying ERα′s post-translational modifications and their impact on cellular physiology and pathology.

I need to use an antibody to detect the protein in samples of equine origin. Do you know if this mouse monoclonal antibody will work?

Rated 2 out of 5 by from Not good for Western blotting, but did work for IPI received a sample of this antibody, which I tested in 2 applications in MCF7 cell lysate: Western blotting and immunoprecipitation. I compared 6 different ERalpha antibodies in my optimization experiments, and this antibody was the only one that did not display any ERalpha (loaded 15 ug lysate) during Western blot visualization. However, when I perform immunoprecipitation with this antibody I see that ERalpha was successfully pulled down.
