EMP3 encodes epithelial membrane protein 3 (EMP-3), a tetraspan-like membrane protein implicated in regulation of cell adhesion, membrane organization, and signal transduction at the cell surface. In human cells, EMP-3 has been linked to modulation of pathways involved in proliferation and stress responses, including receptor-associated signaling and downstream MAPK/PI3K network components depending on cellular context. Altered EMP3 expression patterns have been reported across multiple tumor types and in neurological disease-related studies, supporting its use as a molecular variable in models of oncogenic signaling, invasion, and differentiation. As a membrane-associated factor, EMP-3 is also relevant for dissecting changes in cell–cell interactions and microenvironmental responses.
EMP-3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the EMP3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the EMP3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the EMP3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish EMP-3 protein expression.
This CRISPR knockout system enables efficient generation of EMP3-deficient cell models for investigation of EMP-3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
