    Elastase-1 Antibody (4H5) is a mouse monoclonal IgG2a kappa light chain antibody that detects the Elastase-1 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Elastase-1 antibody (4H5) is available as a non-conjugated format. Elastase-1 is a serine protease that belongs to the elastase subfamily of the peptidase S1 family, primarily secreted by the exocrine pancreas in mammals. However, in humans, Elastase-1 is transcriptionally silent due to mutations that inactivate its promoter and enhancer, which is a significant aspect of its regulation. Elastase-1 plays a crucial role in the digestion of elastin, fibrin, hemoglobin, and albumin, with its activity being inhibited by elafin. Notably, Elastase-1 is expressed exclusively in skin keratinocytes, where Elastase-1 localizes to the basal cell layer of the epidermis, potentially facilitating the detachment of cells from the basement membrane. In keratinocytes, Elastase-1 activity may be modulated by secretory leukocyte protease inhibitor (SLPI) rather than elafin. Furthermore, Elastase-1 has been implicated as a candidate for diffuse nonepidermolytic palmoplantar keratoderma (NEPPK), an autosomal dominant skin condition, highlighting its relevance in dermatological research and potential therapeutic targets.
