DMP-1 Antibody (LFMb-31) is a mouse monoclonal IgG1 kappa light chain antibody that detects DMP-1 protein of human and monkey origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). DMP-1 (LFMb-31) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DMP-1, or dentin matrix protein-1, plays a crucial role in the mineralization of bone and dentin, making it essential for proper skeletal development and maintenance. DMP-1 is predominantly expressed in bone, teeth, and hypertrophic cartilage, where preosteoblasts synthesize this matrix protein. DMP-1 features a unique structure with numerous acidic domains that facilitate function as a transcriptional regulator. DMP-1 localizes to the nucleus of undifferentiated osteoblasts, where DMP-1 activates osteoblast-specific genes and promotes osteoblast differentiation. Upon maturation, DMP-1 undergoes a conformational change and is phosphorylated by casein kinase II in response to calcium influx, followed by export to the extracellular matrix (ECM). Within the ECM, DMP-1 regulates hydroxyapatite nucleation and calcified tissue formation. DMP-1 undergoes proteolytic processing into N- and C-terminal fragments within bone and dentin ECM, and research has identified DMP-1 as a high molecular weight proteoglycan composed of the N-terminal fragment and chondroitin sulfate. Loss of DMP-1 can lead to hypomineralized bone, highlighting DMP-1′s importance in skeletal health.
