Dio-1 Antibody (B-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects Dio-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Dio-1 (B-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Dio-1, also known as Death Inducer-Obliterator-1, is a putative transcription factor characterized by two zinc finger motifs, which are crucial for DNA-binding capability. Dio-1 plays a significant role in regulating apoptosis, particularly during limb development, where Dio-1 translocates to the nucleus to activate programmed cell death. The precise regulation of apoptosis is vital for shaping and defining digits, as apoptosis determines tissue presence. Dio-1 expression increases in response to apoptotic signals, leading to apoptosis induction, a process similarly influenced by NFκB and nerve growth factor expression in reaction to external stimuli. Additionally, caspase inhibitors and Bcl-2 expression modulate Dio-1 activity, reinforcing Dio-1′s critical function in initiating programmed cell death.
