DEC-205 Antibody (F-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects DEC-205 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-DEC-205 antibody (F-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DEC-205, also known as LY75 or lymphocyte antigen 75, is a 1,695-residue multilectin receptor that plays a crucial role in the immune system by mediating the uptake of antigens and their presentation to T cells. DEC-205 is primarily expressed in mature dendritic cells, where DEC-205 facilitates the targeting of membrane receptors to endosomes or lysosomes that are rich in major histocompatibility complex class II (MHC II) products, thereby enhancing the immune response. The structure of DEC-205 includes an extracellular N-terminal cysteine-rich domain, a fibronectin type II domain, ten C-type carbohydrate recognition domains, a single transmembrane region, and a small cytoplasmic C-terminal domain that contains a critical tyrosine residue at position 1679. This structural configuration is essential for DEC-205 function, as DEC-205 can elicit either an agonistic or antagonistic effect on interleukin-4 (IL-4) signaling, influencing the maturation of epithelial cells and the proliferation of T cells. DEC-205 monoclonal antibody (F-4) versatility in various applications makes DEC-205 monoclonal antibody (F-4) an invaluable tool for researchers studying immune responses and dendritic cell biology.

I want to make an CO-IP (co-immunoprécipitation) experement with anti-ly75 (F-4) in non-denaturing conditions in order to detect the binding partner of my target. Does this antibody recognize the native protein or just the denatured form?

Asked by: Smehdi
Thank you for your question. We have not yet tested this antibody, DEC-205 (F-4): sc-515016, for this application and do not know if the antibody has an epitope that will recognize both the native and denatured forms of the antigen. For a limited time, we are offering a free 10 µg sample of this antibody. Please contact our Technical Service Department if you would like to test the antibody before placing your order. Please call us toll-free at (800) 457-3801 option 2, e-mail <scbt@scbt.com>, or use the live chat on our website.

What blocking solution should I use with DEC-205 (F-4): sc-515016 monoclonal antibody for Western blot?

Asked by: Cweed
Thank you for your question. We recommend blocking with Blotto (either TBS Blotto A: sc-2333 or TBS Blotto B: sc-2335). Our Western blot protocol can be found here: https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting

Rated 5 out of 5 by from Excellent antibody I recommend itThe antibody works very well at a 1/100 dilution in ovarian cancer cells. detects both dec205 and fusion proteins : dec205-cd302 at about 210 to 230kda. I found, however, a strong non specific band at 150kda.
