SEE ALSO...

DC-SIGN Antibody (DC28) is a mouse monoclonal IgG2a kappa light chain antibody that detects DC-SIGN protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). DC-SIGN (DC28) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DC-SIGN, also known as CD209, is a type II membrane protein predominantly expressed on dendritic cells (DCs), which are crucial antigen-presenting cells in the immune system. These cells are strategically located in peripheral mucosal tissues and migrate to lymphoid organs, playing a vital role in initiating immune responses. DC-SIGN is essential for mediating the interaction between dendritic cells and resting T cells, as DC-SIGN binds to ICAM-3, facilitating the formation of the immunological synapse. This interaction is particularly significant in HIV-1 infection, as dendritic cells capture the virus through DC-SIGN, promoting viral transport from mucosal surfaces to lymphoid tissues, where HIV-1 can efficiently infect T cells. Understanding DC-SIGN function is critical for developing therapeutic strategies against HIV-1 and enhancing immune responses, making anti-DC-SIGN antibody (DC28) an invaluable tool for researchers studying immune interactions and viral pathogenesis.

I am using DC-SIGN (DC28): sc-65740 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Randy McDonald
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
