DAGLβ Antibody (A-5) is a mouse monoclonal IgG1 antibody that detects the DAGL beta protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). DAGLβ Antibody (A-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DAGLβ, also known as diacylglycerol lipase beta or KCCR13L, is a 672 amino acid multi-pass membrane protein that plays a pivotal role in lipid metabolism as a member of the AB hydrolase superfamily. DAGLβ is primarily located in the cell membrane, where positioning is crucial for function in catalyzing the hydrolysis of diacylglycerol (DAG) to 2-arachidonoyl-glycerol (2-AG), a key signaling lipid involved in retrograde synaptic signaling and axonal growth. DAGLβ′s membrane localization allows effective interaction with substrate, facilitating rapid signaling responses in neurons. Additionally, DAGLβ operates optimally at a pH of 7 and is sensitive to inhibition by compounds such as p-hydroxy-mercuri-benzoate and mercury chloride, but not by phenylmethylsulfonyl fluoride. Alternative splicing of the DAGLβ gene results in three distinct isoforms, each potentially contributing to diverse regulatory mechanisms of lipid signaling pathways in the nervous system.
