Cytokeratin 16 Antibody (LL025) is a mouse monoclonal IgG1 kappa light chain antibody that detects Cytokeratin 16 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-Cytokeratin 16 antibody (LL025) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Cytokeratin 16 is a member of the cytokeratin family, which consists of a diverse group of intermediate filament proteins crucial for maintaining the structural integrity of epithelial cells. Cytokeratin 16 (LL025) monoclonal antibody primarily targets proteins in the cytoplasm of epithelial cells, particularly in stratified squamous epithelium, where Cytokeratin 16 plays a vital role in providing mechanical support and resilience against physical stress. Cytokeratin 16 serves as a marker for identifying tumor origin in squamous cell carcinoma and can indicate tumor differentiation status. Expression of Cytokeratin 16 occurs in benign stratified squamous epithelium and squamous cell carcinoma of the head and neck, as well as in luminal cells of the mammary gland and sweat ducts, while remaining absent in noninvasive breast carcinomas and normal breast tissue. Mutations in Cytokeratin 16 are associated with several skin disorders, including pachyonychia congenita type 1, nonepidermolytic palmoplantar keratoderma, and unilateral palmoplantar verrucous nevus, highlighting Cytokeratin 16′s significance in both normal physiology and disease states.

I am using Cytokeratin 16 (LL025): sc-53255 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: jerojero
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
