CYP27A1 Antibody (G-2) is a mouse monoclonal IgG2a kappa light chain antibody that detects CYP27A1 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). CYP27A1 (G-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CYP27A1 plays a crucial role in cholesterol metabolism and is involved in the synthesis of bile acids, which are essential for the digestion and absorption of dietary fats. CYP27A1 catalyzes the conversion of cholesterol to bile acids, thereby regulating cholesterol levels in the body and influencing lipid metabolism. Dysregulation of CYP27A1 can lead to metabolic disorders, highlighting CYP27A1′s importance in maintaining homeostasis. CYP27A1 is localized to the mitochondria in the liver and kidney, where CYP27A1 performs enzymatic functions, making cellular location vital for activity and overall metabolic processes.

Application: Western blot Sample: whole lysate of human hepatocytes from Huh7 cell line Gel running conditions: reduced, denaturing, 10% SDS-PAGE Loading amount: 30 µg protein/lane Transfer: semi-dry, on nitrocellulose membrane (0.45 µm) Blocking agent: 5% milk in TBST buffer, for 1h, RT Primary antibody dilution- 1: 500 in 5% milk, for 16 h at 4°C Secondary antibody: non- Santa Cruz antibody anti-mouse-HRP, 1:10000 dilution in 5% milk, for 1h, RT Detection: ECL substrate (femto sensitivity) Exposure time: 0.5 sec Bands: specific, at approx.55 kDa; non-specific bands are absent.

Rated 5 out of 5 by from Cyp27a1 antibody gives good bands on Liver WesternReceived a Cyp27a1 sample antibody with a Cyp8b1 sample antibody for the revision of one of my manuscripts. And both worked really well with mouse liver samples on westerns. I loaded 20-50mcg protein, used 1:1000 dilution of primary (Cyp27a1 and Cyp8b1 respectively) and then secondary antibody each, and developed with a "femto" enhancer instead of a "pico" (=regular enhancer) which I commonly do, and I got nice strong bands. I know many people use lower concentrations of primary antibody (which should work as well I think); but we tend to use these concentrations in our lab. Bottom line: This Cyp27a1 antibody works well, and I am happy that it did in my case, as I had several other experiments to complete for my revision.
