CXCR-4 Antibody (12G5) is a mouse monoclonal IgG2a kappa light chain antibody raised against living CP-MAC-infected SUP-T1 cells, specifically targeting the CXCR-4 protein of human origin, and is validated for applications including immunoprecipitation (IP) and flow cytometry (FCM). CXCR-4, a member of the C-X-C chemokine receptor family, is a G protein-coupled receptor characterized by its seven transmembrane domains and its interaction with stromal cell-derived factor-1 (SDF-1). This receptor plays a pivotal role in cellular signaling and migration, acting as a potent chemoattractant for T cells and monocytes. The CXCR-4/SDF-1 axis is critical for numerous physiological processes, including hematopoiesis, immune response, and organogenesis, as well as pathological conditions like cancer metastasis and immune disorders. CXCR-4 is also the primary co-receptor for T-tropic HIV-1, facilitating viral entry into target cells, while its ligand SDF-1 has been shown to inhibit HIV-1 infection, making this receptor-ligand pair an important focus of antiviral and immunological research. Highly homologous to other IL-8 receptors, CXCR-4 shares approximately 37% sequence identity at the amino acid level, yet exhibits distinct ligand specificity and functional roles. CXCR-4 monoclonal antibody (12G5) is available in non-conjugated form as well as a range of conjugated options, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and Alexa Fluor® conjugates, offering flexibility for diverse research applications.

what is the difference between "sc-12764" L and "sc-12764"? is only the concentration?

Thank you for your question. The regular version of CXCR-4 Antibody (12G5): sc-12764 is supplied as 200 µg in 1 ml PBS with 0.1 % gelatin and <0.1 % sodium azide. The L version is supplied in concentrated formulation at 200 µg antibody in 0.1 ml PBS with no gelatin or sodium azide.

I am looking for blocking peptide for this antibody CXCR-4 (12G5). Please suggest to inhibit antibody binding with target.

Thank you for your question. CXCR-4 Antibody (12G5) is a mouse monoclonal IgG2a raised against living CP-MAC-infected SUP-T1 cells of CXCR-4 of human origin. The antibody is not raised against a synthetic peptide, and therefore no blocking peptide is available. An alternative negative control would be to use an isotype control in lieu of the CSCR-4 specific antibody. We would recommend our normal mouse IgG 2a: sc-3878.

Can CXCR-4 (12G5): sc-12764 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, CXCR-4 (12G5): sc-12764 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
