Them4 encodes CTMP (carboxyl-terminal modulator protein), a membrane-associated regulator best known for modulating AKT/PKB signaling. CTMP can bind AKT and influence its phosphorylation state, linking Them4 to PI3K–AKT pathway control of cell survival, metabolism, and stress responses. In mouse systems, altered CTMP activity has been associated with changes in mitochondrial function, oxidative stress handling, and apoptosis-related processes. Because PI3K–AKT signaling is broadly implicated in inflammatory, metabolic, and neurodegenerative phenotypes, Them4 is frequently studied as a node connecting signaling dynamics to disease-relevant cellular states.
CTMP CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Them4 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Them4 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Them4 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish CTMP protein expression.
This CRISPR knockout system enables efficient generation of Them4-deficient cell models for investigation of CTMP signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
