CRMP-4 Antibody (LL-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects CRMP-4 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). CRMP-4 (LL-5) antibody is available as a non-conjugated format. Collapsin response mediator proteins (CRMPs), including CRMP-1, CRMP-2, CRMP-3, CRMP-4, and CRMP-5, play a crucial role in neuronal signaling, particularly in response to the axon guidance molecule semaphorin 3A (SEMA3A). CRMP-4 protein is primarily located in the developing cerebral cortex and neocortical neurons, where CRMP-4 is essential for proper neuronal differentiation and plasticity. Notably, CRMP-4 is expressed in the perikaryon, neurites, and growth cones of rodent neocortical neurons, highlighting CRMP-4′s importance in neuronal development and connectivity. In the adult brain, while CRMP expression is significantly reduced, CRMP-4 continues to be present in regions such as the dentate gyrus of the hippocampus, which is known for neurogenesis and synaptic plasticity. This persistent expression in specific brain regions underscores CRMP-4′s significance in maintaining neural plasticity and adaptability throughout life.

This antibody has worked well for us for Western Blot. I now want to test CRMP4 levels by immunofluorescence in mouse brain slices. This antibody is only recommended for WB, is there any chance it would work for IF? Or would that be a waste of time?

Thank you for your question. We have not yet tested this antibody for IF or IHC in tissues or cells, and so this is not a recommended application at this time. However, it certainly may be worth trying since you already have the antibody. The two citations we are aware of currently are using the antibody in Western blot and IP/WB.

For Western Blot, is it recommended to use denatured or non-denatured conditions with CRMP-4 (LL-5): sc-100323 antibody?
