Cox-2 Antibody (H-3) is a mouse monoclonal IgM kappa antibody specific for an epitope mapping between amino acids 17-55 near the N-terminus of Cyclooxygenase-2 (Cox-2) of rat origin; anti-Cox-2 antibody (H-3) reacts with mouse, rat, and human proteins and is suitable for applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), enzyme-linked immunosorbent assay (ELISA), and immunohistochemistry. Cox-2, also known as Prostaglandin-Endoperoxide Synthase 2 (PTGS2), is an inducible enzyme that plays a crucial role in prostaglandin biosynthesis from arachidonic acid, mediating processes like inflammation, pain, and fever. Structurally, Cox-2 is a homodimeric enzyme with each subunit containing a heme prosthetic group essential for catalytic activity; Cox-2 possesses both cyclooxygenase and peroxidase active sites within a single polypeptide chain. The structural differences between Cox-2 and Cox-1 isoform, such as a larger substrate channel and a distinct hydrophobic pocket, allow for selective inhibition by certain nonsteroidal anti-inflammatory drugs (NSAIDs), minimizing gastrointestinal side effects associated with non-selective NSAIDs. Understanding Cox-2 structure is important for developing selective inhibitors that can effectively target inflammatory pathways without affecting physiological functions mediated by Cox-1. Cox-2 overexpression has been implicated in various pathological conditions, including chronic inflammation and tumor progression, making Cox-2 a valuable target for therapeutic intervention and research.
