    Complexin-1/2 Antibody (D-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects Complexin-1/2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). anti-Complexin-1/2 antibody (D-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Complexin-1 and Complexin-2, also known as Synaphin-1 and Synaphin-2, are critical proteins that contain an alpha-helical middle domain of approximately 58 amino acids, which is essential for synaptic transmission. These proteins are predominantly expressed in presynaptic terminals of both inhibitory and excitatory neurons in the hippocampus, as well as in cytoplasmic pools during early developmental stages. Complexin-1 and Complexin-2 are vital for regulating neurotransmitter release, as these proteins facilitate SNARE (soluble N-ethylmaleimide-sensitive factor attachment protein receptors) precomplex formation by binding to syntaxin through their alpha-helical domain. This interaction enables calcium-dependent release of neurotransmitters, as neurons lacking complexins show reduced transmitter release efficiency due to diminished calcium sensitivity in synaptic secretion. Additionally, Complexin-2 plays a role in long-term potentiation following tetanic stimulation, contributing to synaptic plasticity. A progressive loss of Complexin-2 has been observed in brains of mice with the Huntington disease mutation, suggesting that altered neurotransmitter release may contribute to motor, emotional, and cognitive dysfunctions associated with this disorder.

    Thank you for your question. Yes, Complexin-1/2 (D-9): sc-365152 has been validated for use in IHC with paraffin-embedded sections.
