Clusterin Antibody (CLI-9) is a mouse monoclonal IgG1 antibody designed to recognize full-length native Clusterin of human origin, and is recommended for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) applications. Clusterin, also known as apolipoprotein J (ApoJ) or complement lysis inhibitor (CLI), is a multifunctional secretory heterodimeric glycoprotein composed of α and β subunits linked by disulfide bonds, resulting from the post-translational cleavage of a 449 amino acid precursor. This intricate structure enables Clusterin to participate in a variety of critical physiological processes, including immune regulation, cell adhesion, lipid transportation, tissue remodeling, membrane recycling, and cell-cell interactions. One of the key functions of Clusterin is its role as a complement lysis inhibitor, where Clusterin interacts with complement proteins such as C3 and C5 to prevent the formation of the membrane attack complex, thereby protecting cells from complement-mediated lysis and maintaining immune homeostasis. Additionally, Clusterin interacts with apoptotic proteins like Bax, modulating apoptosis and promoting cell survival under stress conditions. In the context of neurodegenerative diseases, Clusterin binds to amyloid-beta peptides, influencing their aggregation and reducing neuritic toxicity, which is significant in the pathogenesis of Alzheimer′s disease. Furthermore, Clusterin interacts with various extracellular matrix proteins, facilitating cell adhesion and migration, which are essential processes in tissue remodeling and cancer metastasis. Overexpression of Clusterin has been observed in advanced stages of several cancers, including mammary tumors, where Clusterin supports tumor progression and metastasis by enhancing cell survival and resistance to apoptosis. Anti-Clusterin antibody (CLI-9) enables detailed studies of Clusterin′s interactions with these critical proteins, providing valuable insights into its role in immune regulation, apoptosis, neuroprotection, and cancer biology.
