CLIP-170 Antibody (F-3) is a mouse monoclonal IgG2b kappa light chain antibody that detects CLIP-170 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). CLIP-170 (F-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The cytoplasmic linker protein (CLIP-170) plays a crucial role in cellular dynamics as a member of the plus-end-binding proteins (+TIPs), which are essential for microtubule stabilization and organization. By binding to the growing plus ends of microtubules, CLIP-170 acts as a linker between these dynamic structures and organelle membranes, thereby promoting microtubule rescue and preventing catastrophe, particularly near the cell periphery. This function is vital for maintaining cellular integrity and facilitating intracellular transport, as CLIP-170 ensures that microtubules remain stable and functional during cellular processes such as mitosis and organelle movement. Additionally, fluorescently labeled CLIP-170 can be visualized as comet-like streaks at the growing ends of microtubules, providing insights into dynamic behavior in live-cell imaging studies. CLIP-170 also co-localizes with Dynactin and Dynein at microtubule ends and at the kinetochore, highlighting CLIP-170′s importance in the orchestration of cellular transport and division. Furthermore, CLIP-170 is highly expressed in Hodgkin and Reed-Sternberg (HRS) cells, monocyte-derived dendritic cells, interleukin-4 plus CD40 ligand activated B cells, and Ki-1 lymphoma, making CLIP-170 a significant marker in various pathological conditions.

Can CLIP-170 (F-3): sc-28325 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Asked by: Dr Ninau Qelp
Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, CLIP-170 (F-3): sc-28325 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
