Claudin-1, also known as CLDN1, is a key member of the claudin family, essential for forming tight junctions that regulate paracellular transport in epithelial and endothelial cells. Located at the plasma membrane, CLDN1 spans the membrane four times, creating a robust barrier that separates distinct cellular environments. This localization is crucial, as CLDN1′s role in tight junctions supports cell polarity and tissue integrity, which is particularly important in organs like the kidney and liver where high expression levels aid in selective permeability. Additionally, CLDN1 interacts with other tight junction proteins, such as occludins and junctional adhesion molecules, to form tight junction strands that are critical for compartmentalizing tissue spaces. Variations or defects in CLDN1 expression have been implicated in conditions like neonatal ichthyosis-sclerosing cholangitis syndrome, underscoring its role in cellular barrier function. Claudin-1 (D-4) antibody is available in non-conjugated and various conjugated formats, making it versatile for detailed study of this vital protein across species.
