CKR-5 Antibody (D-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects CKR-5 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). CKR-5 (D-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CKR-5 protein, also known as CCR5, is a member of the C-C chemokine receptor family, which plays a crucial role in immune response by mediating chemotaxis of T cells and monocytes to sites of inflammation. CKR-5 serves as a co-receptor for HIV, facilitating viral entry into host cells, particularly in macrophage-tropic strains of the virus. This interaction underscores CKR-5′s significance in both normal immune function and HIV infection pathology, making CKR-5 a critical target for therapeutic interventions aimed at blocking viral entry. CKR-5′s structure, characterized by seven transmembrane domains, allows effective signal transmission in response to chemokine binding, which is essential for orchestrating immune responses. Using anti-CKR-5 antibody (D-6) in research can provide valuable insights into immune cell trafficking mechanisms and CKR-5′s role in viral pathogenesis.

What are the exact conditions used to obtain the Western blot data shown on your website for CKR-5 (D-6): sc-17833 monoclonal antibody?

Asked by: cjMara
Thank you for your question. We used this primary antibody at a dilution of 1:200 and incubated for one hour at room temperature in Blotto (sc-2333). The complete recommended protocol for Western blotting is provided below. https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting

Rated 1 out of 5 by from anti-CKR5 Ab is not working (very non-specific)Initially, I was having trouble determining the right band for CCR5 on the WB from all the other non-specific bands. Therefore, I decided to run true negative control. I used tzm-bl cell (positive for CCR5 expression) and Hela cells (negative control) to chekc any differences in banding pattern and I do not see any difference. There should not be any CCR5 band in Hela cells.

Rated 4 out of 5 by from Clear with exact positionDiluted 1:500 with casein buffer, samples from ACHN cell line, PVDF memberane with 300mA 120min transfering. It is easy to scan with Odyssey system. The CKR5 is located at about 46 kDa, however, there are two non-specific bands above it.

Rated 3 out of 5 by from some non-specific banduse the monoclone antibody with dilution of 1:500, I can detect the CCR5 band successfully, but there are still some non-specific band on the membrane.
