CIP2A Antibody (2G10-3B5) is a mouse monoclonal IgG2b kappa light chain antibody that detects CIP2A of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. CIP2A (2G10-3B5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The cancerous inhibitor of protein phosphatase 2A (CIP2A), also known as p90 autoantigen or KIAA1524, is a single-pass membrane protein that plays a critical role in oncogenesis by inhibiting protein phosphatase 2A (PP2A) activity. PP2A inhibition is significant as this vital tumor suppressor regulates various cellular processes, including cell growth and division. By preventing PP2A from dephosphorylating c-Myc, an oncogenic transcription factor, CIP2A stabilizes c-Myc, thereby promoting tumor formation and malignant cell proliferation. CIP2A interaction with PP2A′s catalytic subunit is crucial for human cell transformation progression, highlighting CIP2A′s importance in cancer biology. CIP2A overexpression occurs frequently in several cancer types, including colon, gastric, and head and neck squamous cell carcinomas, underscoring CIP2A′s potential as a therapeutic target and biomarker for cancer diagnosis and prognosis.

The antibody works beautifully on immunofluorescence and immunohistochemistry for cells. However, it does not work on ANY FFPE tissues. We have tried various antigen retrieval approaches but that doesn't seem to work either.

Hello, I would like to ask you for a more detailed description of the conditions for IHC-P with SC-80659 antibody (antigen retrieval, concentration of antibody, incubation time). Well thank you

Thank you for your question. For this antibody we recommend a starting dilution of 1:50. We do recommend antigen retrieval with sodium citrate and heat. Incubation time needs to be optimized for your samples, but I would start with an overnight primary incubation, and taper that down if needed. Can you recommend a positive control to use with CIP2A (2G10-3B5): sc-80659 monoclonal antibody for Western blot?

We recommend HeLa (sc-2200), SW480 (sc-2219), COLO 320DM (sc-2226) or Caco-2 (sc-2262) whole cell lysates to use as positive controls as we have great Western blot data showing specific detection of CIP2A in these cell lines.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with CIP2A (2G10-3B5): sc-80659?

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. Especially in mouse tissues many antibodies crossreact with an unknown 90 kD protein This antibody we have validated to be specific by using lysates from CIP2A KO MEFs (HOZ =KO). This is also the antobody we have found to work best for IHC of human tumor tissues after rigorous testing
