Cingulin Antibody (G-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects human Cingulin protein through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Cingulin monoclonal antibody (G-6) is available in non-conjugated form and conjugated with agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® variants. Cingulin plays a crucial role in maintaining tight junction integrity, which regulates paracellular permeability and cell polarity. Located at the submembrane plaque of tight junctions, Cingulin interacts with other key proteins, including PDZ protein ZO-1, to facilitate junction assembly and stability. Cingulin′s structural composition, featuring globular and coiled-coil domains, enables interactions with cytoskeletal proteins and tight junction components. Cingulin inhibits RhoA activation, a critical signaling pathway involved in cytoskeletal dynamics, influencing cell shape and motility. Increased Cingulin expression in confluent cells leads to RhoA signaling downregulation by inhibiting guanine nucleotide exchange factors, highlighting its importance in cellular signaling and junctional integrity.
