CD9 Antibody (KMC8.8) is a rat monoclonal IgG2a antibody raised against BMS2 murine bone marrow stromal cells, reactive with mouse species, and validated for applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), flow cytometry (FCM), and immunohistochemistry. CD9, a member of the tetraspanin family, is a type IV transmembrane glycoprotein characterized by its four transmembrane domains and two extracellular loops, a structural configuration that is critical for its biological functions. This distinctive structure facilitates the formation of tetraspanin-enriched microdomains (TEMs) on the cell surface, which serve as platforms for protein-protein interactions and cellular signaling. CD9 is widely expressed on the surface of various cell types, including pre-B cells, eosinophils, basophils, activated T cells, and platelets, and plays a crucial role in mediating cell adhesion, migration, and intercellular communication. CD9′s structural properties enable association with integrins and other transmembrane proteins, influencing signal transduction pathways essential for processes such as immune response, tissue regeneration, and hemostasis. In platelets, CD9 is a major component of the cell surface and contributes to aggregation and clot formation, underscoring CD9′s importance in vascular biology. CD9 is also implicated in hematological malignancies, being expressed on most non-T acute lymphoblastic leukemia cells and some acute myeloid and chronic lymphoid leukemias, making CD9 a valuable marker in diagnostic and therapeutic research. Anti-CD9 antibody (KMC8.8) is available in non-conjugated and conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and Alexa Fluor® options, providing researchers with a versatile tool for in-depth exploration of CD9′s structure-function relationships and roles in cellular and pathological contexts.
