CD8A encodes CD8-α, a type I membrane glycoprotein that forms CD8αα homodimers or CD8αβ heterodimers on T cells and a subset of NK cells, acting as a co-receptor for MHC class I–restricted antigen recognition. By binding MHC I and associating with LCK, CD8-α promotes TCR signaling, immunological synapse formation, and downstream pathways controlling cytotoxic effector differentiation and cytokine production. CD8A expression is widely used to define cytotoxic lymphocyte states and to dissect lineage specification, activation, and exhaustion programs. Dysregulation of CD8-α–dependent signaling and CD8+ T-cell phenotypes is relevant to tumor immune microenvironments, chronic viral infection, autoimmunity, and immune deficiency research contexts.
CD8-α CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the CD8A gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the CD8A together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the CD8A open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish CD8-α protein expression.
This CRISPR knockout system enables efficient generation of CD8A-deficient cell models for investigation of CD8-α signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
