CD5 Antibody (D-9) is a mouse monoclonal IgG1 antibody that detects CD5 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). CD5 is a crucial transmembrane glycoprotein predominantly expressed on T lymphocytes, accounting for approximately 70% of normal peripheral blood lymphocytes and nearly all T cells in the thymus and peripheral blood. CD5 plays a significant role in modulating T cell activation; when T cells are activated via the T cell receptor (TCR), CD5 undergoes tyrosine phosphorylation, which is essential for regulating T cell responses. Notably, CD5 absence leads to hyper-responsiveness of T cells to TCR-mediated activation, highlighting CD5′s importance in maintaining immune homeostasis. CD5 interacts with the TCR/CD3-ζ chain and the Src family kinase Lck, enhancing Lck′s kinase activity by 10- to 15-fold when bound to CD5. Additionally, CD5 serves as a receptor for the B cell antigen CD72, although the precise consequences of this interaction remain to be fully elucidated, suggesting a potential role in thymic selection processes.
