CD45RB Antibody (16A) is a rat monoclonal IgG2a antibody engineered to target the CD45RB isoform of the CD45 protein, also known as Protein Tyrosine Phosphatase Receptor Type C (PTPRC). CD45RB is a vital transmembrane glycoprotein expressed on the surface of all nucleated hematopoietic cells in mice, playing a crucial role in regulating immune cell signaling and activation. Structurally, CD45RB consists of an extracellular domain generated through alternative splicing of exons 4, 5, and 6, a single transmembrane region, and a conserved intracellular domain that possesses phosphatase activity. This structural organization is essential for CD45RB′s function, as the variability in the extracellular domain allows for differential interactions with other cell surface molecules, thereby modulating signal transduction pathways critical for T and B cell receptor signaling. Located on the cell membrane, CD45RB dephosphorylates key tyrosine residues on Src family kinases such as Lck and Fyn, as well as on Syk family kinases like ZAP-70, which are essential for initiating and sustaining signaling cascades that drive lymphocyte development, activation, and immune responses. Additionally, CD45RB undergoes significant post-translational modifications, including extensive glycosylation, which are crucial for its stability, proper localization on the cell membrane, and interactions with other signaling proteins, thereby enhancing its regulatory functions. Anti-CD45RB antibody (16A) is raised against clone TH2 mouse cells expressing CD45RB and is effective in various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM) across mouse, rat, and human samples. CD45RB monoclonal antibody (16A) facilitates comprehensive analysis of immune cell populations and their interactions with key signaling proteins, providing valuable insights into immune system dynamics and the mechanisms underlying immune-related diseases. CD45RB (16A) antibody′s ability to detect multiple isoforms of CD45RB, resulting from alternative splicing, allows researchers to investigate the diversity and function of different immune cell subsets, making CD45RB (16A) monoclonal antibody an indispensable tool for both research and diagnostic purposes in immunology and hematology.

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to phycoerythrin (sc-18846 PE) or fluoroscein (sc-18846 FITC).
