CD45 Antibody (2D-1) is a mouse monoclonal IgG1 kappa light chain antibody engineered to detect CD45, also known as Protein Tyrosine Phosphatase Receptor Type C (PTPRC), in mouse, rat, and human samples using western blotting (WB), immunoprecipitation (IP), PAR assays, and flow cytometry (FCM). CD45 monoclonal antibody (2D-1) targets the extracellular domain of human CD45. CD45 is a vital transmembrane glycoprotein prominently expressed on the surface of all hematopoietic cells, where CD45 plays a crucial role in regulating immune cell signaling and activation. Located on the cell membrane, CD45 acts as a key regulator of T and B cell receptor signaling by dephosphorylating specific tyrosine residues on Src and Syk family kinases, such as Lck, Fyn, and ZAP-70. This modulation is essential for proper immune responses, including T cell activation, proliferation, and differentiation, as well as B cell development and antibody production. The extracellular domain of CD45 undergoes alternative splicing, resulting in multiple isoforms that provide fine-tuned regulation of immune functions tailored to different cell types and activation states. The precise localization of CD45 on the cell surface facilitates CD45 interaction with other membrane-bound receptors and signaling molecules, making CD45 indispensable for maintaining immune homeostasis and responding to pathogens. Dysregulation or altered expression of CD45 has been implicated in various immune disorders, including autoimmune diseases and leukemias, highlighting CD45 significance in both normal immune function and disease pathogenesis. Anti-CD45 antibody (2D-1) is available in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and Alexa Fluor® conjugates, providing versatile options for a wide range of experimental applications. By utilizing CD45 monoclonal antibody (2D-1), researchers can effectively study the expression, isoform distribution, and functional dynamics of CD45, thereby advancing our understanding of immune cell biology and related disease mechanisms.

Is there any cd45 antibody can use in hamster?

Thank you for your question. Unfortunately, none of our CD45 antibodies are currently recommended for use in hamster, nor are we aware of any product citations using the CD45 antibodies in hamster. This monoclonal antibody, CD45 Antibody (B-8): sc-28369, is raised against a recombinant protein (amino acids 1075-1304 of CD45 of human origin) which shares 75% sequence identity with this published model hamster CD45 protein sequence, (accession number: XP_012971285). The exact epitope recognized by this monoclonal antibody has not been determined, however, based on this alignment there is a chance that the antibody may cross-react with hamster. However, because the alignment is not 100%, we still can not guarantee it.

Can CD45 (2D-1): sc-1187 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, CD45 (2D-1): sc-1187 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

The only reason why I am not giving a 5-stars is because the intensity of the staining was not very strong. I was trying to stain lymphocytes infiltration in human lung sections.
