SEE ALSO...

CD42d Antibody (G-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects CD42d protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). CD42d (G-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CD42d plays a crucial role in the early phase of primary hemostasis, where CD42d functions as part of the CD42 complex, also known as the platelet glycoprotein complex. This complex facilitates platelet adhesion to damaged blood vessel walls by binding to the von Willebrand factor, which becomes exposed on the subendothelium during vascular injury. The CD42 complex consists of four subunits: CD42b (GPIb alpha), CD42c (GPIb beta), CD42a (GPIX), and CD42d (GPV), with CD42a and CD42d being noncovalently linked to the complex. CD42d is specifically expressed in platelets and megakaryocytes, and CD42d cleavage by thrombin results in both a soluble fragment and a membrane-associated fragment, making CD42d an excellent marker for platelet activation. The gene encoding human CD42d is located on chromosome 3q29, highlighting CD42d significance in hemostatic processes and potential implications in thrombotic disorders.

Merci.

I am using CD42d (G-11): sc-271662 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Randy McDonald
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
