Mouse Cd2 encodes CD2, an immunoglobulin superfamily adhesion and signaling receptor expressed predominantly on T cells and NK cells that mediates cell–cell contact through interaction with ligands such as CD58/CD48. CD2 participates in immunological synapse formation and co-stimulatory signaling that shapes T cell activation thresholds, cytoskeletal remodeling, and downstream pathways including Src family kinase, MAPK, and NF-κB signaling. By influencing lymphocyte adhesion, migration, and effector differentiation, CD2 contributes to the regulation of adaptive immune responses and immune homeostasis. Dysregulated CD2-dependent signaling and adhesion have been implicated in inflammatory and autoimmune-associated immune phenotypes, supporting its use in mechanistic studies of T cell function.
CD2 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Cd2 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Cd2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Cd2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish CD2 protein expression.
This CRISPR knockout system enables efficient generation of Cd2-deficient cell models for investigation of CD2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
