CD163 Antibody (RM3/1) is a mouse monoclonal IgG1 kappa light chain antibody designed to detect human CD163 with exceptional precision through immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). CD163, also known as M130 or SCARI1, is a pivotal macrophage-associated antigen within the scavenger receptor cysteine-rich (SRCR) superfamily, predominantly localized on the surface of macrophages and, to a lesser extent, on monocytes. This cell surface localization enables CD163 to function effectively as a receptor that mediates the scavenging and endocytosis of haptoglobin-hemoglobin complexes, thereby playing a significant role in anti-inflammatory response and iron metabolism. The membrane-bound position of CD163 facilitates interaction with extracellular haptoglobin-hemoglobin, preventing oxidative stress and maintaining cellular homeostasis during hemolytic events. Anti-CD163 antibody (RM3/1) is available in both non-conjugated form and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatile options for diverse experimental applications. The receptor-ligand binding of CD163 is calcium-dependent and exhibits high affinity, and multiple CD163 isoforms, which vary in cytoplasmic domains and potential phosphorylation sites, suggest distinct signaling mechanisms that regulate macrophage function. The gene encoding CD163 is located on human chromosome 12p13.31, highlighting genetic significance and potential implications in various inflammatory and immune-mediated diseases. This comprehensive characterization of CD163 underscores importance in cellular processes and validates CD163 (RM3/1) antibody as an essential tool for researchers studying macrophage biology and related pathways.
