CBP Antibody (C-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects CBP protein of mouse, rat, and human origin through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry (IHC), and enzyme-linked immunosorbent assay (ELISA). CBP (C-1) antibody is offered in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and various Alexa Fluor® conjugates. CBP protein, also known as CREBBP or KAT3A, plays a crucial role in regulating gene expression in response to cyclic AMP signaling, particularly through interaction with the cAMP-regulated enhancer (CRE). This enhancer is a key regulatory element bound by several transcription factors, including CREB, which is activated by phosphorylation from protein kinase A. Binding of phosphorylated CREB to CBP is essential for transcriptional co-activation of target genes, highlighting CBP importance in mediating cellular responses to hormonal signals. CBP shares significant homology with another transcriptional co-activator, p300, suggesting both proteins are part of a conserved family that plays vital roles in gene regulation. CBP interaction with various transcription factors and involvement in critical cellular processes underscore significance in molecular biology and potential implications in understanding diseases related to gene expression dysregulation.

I recently purchased this antibody. However, I need to know if this antibody cross-reacts with the closely related p300 protein (KAT3B aka EP300) in human cells. A rapid and accurate response would be greatly appreciated. Thanks, Isaac

Thank you for your question. This mouse monoclonal antibody, CBP (C-1): sc-7300, is specific for CBP (CREB-binding protein). We have not specifically tested this clone for potential cross-reactivity with KAT3B aka EP300, however this antibody has been validated for specificity using our human siRNA for knockdown against the human gene target, CBP siRNA (h): sc-29244.

Is there any data showing this antibody is specific to CBP?

Thank you for your question. We have immunofluorescence data with methanol fixed HeLa cells treated with siRNA to show knockdown of signal, confirming specificity to CBP.

Rated 1 out of 5 by from not working for me in IFAI was unfortunately not successful in getting a signal with 96 well plate probed with recombinant CBP tagged protein O/N at 4C using a fluorescent secondary antibody for 1hr. Antibodies were concentrated as high as 1:50 in PBS-BSA and incubated for 1-2hrs at RT. I used an anti-CBP from a different vendor that gave me a specific signal. This was also tried on fixed cell lines expressing CBP on the surface with negative results as well.
