Cathepsin W Antibody (E-9) is a mouse monoclonal IgG1 light chain antibody that detects cathepsin W in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Cathepsin W is a member of a unique subgroup of cathepsin proteases, specifically expressed in CD8+ T lymphocytes, and plays a crucial role in the immune response by regulating the activation and differentiation of these cells. Cathepsin W is initially expressed during the transition of thyrocytes to CD8+ T lymphocytes, coinciding with the downregulation of CD4+ receptors in thymocytes, highlighting its importance in T cell development. Cathepsin W is localized within the rough endoplasmic reticulum in transfected Cos-7 and HeLa cells, which is significant for its processing and maturation. Notably, cathepsin W features a unique 21 amino acid peptide insertion between the active site histidine and asparagine residues, along with a distinctive 8-amino acid carboxy-terminal extension, contributing to its functional specificity. Cathepsin W also exhibits an extended loop structure in the beta-sheet domain and an additional disulfide bond, which are characteristic of its structure. Furthermore, cathepsin W may exist as a dimer, with each monomer linked by a disulfide bond, underscoring its complex nature and potential regulatory mechanisms in immune responses.
