Calponin 1/2/3 Antibody (H-3) is a mouse monoclonal IgG3 antibody that detects Calponin in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Calponin is a crucial actin- and tropomyosin-binding protein that plays a significant role in regulating smooth muscle contraction and serves as a marker for smooth muscle cell differentiation. Calponin primary function is to inhibit smooth muscle actomyosin activity, which is vital for maintaining proper muscle tone and function. Calponin achieves this by interacting with F-Actin and inhibiting the actin-activated MgATPase activity of phosphorylated myosin, thereby modulating contraction dynamics. Notably, Calponin regulatory properties are compromised upon phosphorylation, particularly at Serine 175, by protein kinase C or calmodulin-dependent protein kinase II, which can lead to altered muscle contractility. The three isoforms—Calponin 1 (basic Calponin), Calponin 2 (neutral Calponin), and Calponin 3 (acidic Calponin)—are predominantly expressed in smooth muscle tissue, with Calponin 2 also present in heart muscle and Calponin 3 found in the brain, highlighting the diverse roles across different tissues.
