SEE ALSO...

CALML5 Antibody (A-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects CALML5 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). CALML5 (A-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CALML5, also known as calmodulin-like 5 or CLSP, is a 146 amino acid protein characterized by four EF-hand domains, which are critical for calcium binding. This structural feature enables CALML5 to function similarly to calmodulin, acting as a calcium sensor that modulates various cellular processes. CALML5 is predominantly expressed in the epidermis, where CALML5 plays a significant role in the terminal differentiation of keratinocytes. CALML5 interacts with transglutaminase 3 (TGase3), a key enzyme involved in the cross-linking of proteins during skin barrier formation, highlighting CALML5′s importance in maintaining skin integrity and function. Through calcium level regulation and protein interactions, CALML5 contributes to vital physiological processes, making CALML5 a valuable target for research in skin biology and related fields.

Why is it in the western blot example of CALML5 (A-3): sc-393637 monoclonal antibody shows the band for Calml5 at 23 kDa, when the mw of Calml5 is approximately 16 - 17 kDa?

Asked by: maxh
Thank you for your question. The blot shown on the product page is of transfected and untransfected 293T whole cell lysates. They have been transfected with a construct encoding a recombinant CALML5 fusion protein, which is not representative of endogenous expression of the protein. This antibody detects endogenous CALML5 at a molecular weight of approximately 16 kDa.

Can CALML5 (A-3): sc-393637 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: AbPolly
Thank you for your question. Yes, CALML5 (A-3): sc-393637 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
