CAF-1 p150 Antibody (D-1) is a mouse monoclonal IgG2b kappa light chain antibody that detects CAF-1 p150 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-CAF-1 p150 antibody (D-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The chromatin assembly factor-1 (CAF-1) complex, which includes the p150 subunit, plays a crucial role in the assembly of nucleosomes by depositing newly synthesized and acetylated histones H3 and H4 into nascent chromatin during DNA replication. CAF-1 p150 (D-1) monoclonal antibody is particularly important for maintaining heterochromatin, a tightly packed form of DNA that is essential for regulating gene expression and ensuring genomic stability. Heterochromatin is characterized by late replication during S phase, low gene density, and presence of repetitive DNA sequences that are less accessible to transcriptional machinery. During late S phase, p150 interacts directly with heterochromatin-associated proteins, such as HP1α, HP1β, and HP1γ, facilitating proper assembly and maintenance of heterochromatin. As cells transition into mitosis, dissociation of CAF-1 p150 and HP1 proteins from heterochromatin occurs in conjunction with phosphorylation of histone H3, highlighting dynamic nature of chromatin organization during cell division. Reassociation of HP1 proteins with chromatin at mitosis end, following dephosphorylation of histone H3, underscores CAF-1 p150′s critical role in chromatin dynamics and cellular function.
