c-Kit Antibody (E-3) is a mouse monoclonal IgG1 kappa light chain antibody specifically engineered to recognize c-Kit protein, a crucial transmembrane receptor tyrosine kinase located on the cell surface. anti-c-Kit antibody (E-3) targets an epitope mapped between amino acids 958-976 within the C-terminus of human c-Kit, ensuring high specificity and reliable detection across mouse, rat, and human species using western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) techniques. c-Kit protein, also known as CD117, plays an essential role in various cellular processes, including cell proliferation, differentiation, survival, and migration, by mediating signaling pathways critical for hematopoiesis, mast cell development, and stem cell maintenance. c-Kit′s strategic location on the cell membrane allows effective interaction with stem cell factor (SCF), facilitating vital communication between cells and their microenvironment, which is fundamental for normal physiological functions and immune responses. Additionally, dysregulation or mutations in c-Kit gene are closely associated with the development and progression of several cancers, such as gastrointestinal stromal tumors and acute myeloid leukemia, highlighting significance as a therapeutic target. c-Kit (E-3) antibody is available in both non-conjugated form and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatile options for researchers to investigate c-Kit′s role in both healthy and diseased states, thereby advancing understanding of contributions to cellular biology and potential in cancer treatment.

Does this antibody work on PFA-fixed brain tissue for IHC?

Thank you for your question. c-Kit Antibody (E-3): sc-365504 is recommended for detection of c-Kit p145 of mouse, rat and human origin by WB, IP, IF and ELISA. We do not recommend using this antibody in IHC applications.

Which antigen retrieval technique is needed for IHC-P ? thanks

This antibody has not yet been tested on formalin-fixed paraffin-embedded tissue sections, or for IHC(P). This antibody is recommended and warrantied for IF (immunofluorescent staining of fresh or frozen tissues or cells fixed in methanol or acetone), however we are still gathering data to show on our website. I tested sample of this antibody (1:400) with mouse thymic epithelial cells and got strong single band. However, the protein band came up around 55~60kda in 8% page gel. I am wondering if this is the right band size for c-kit ?

Thank you for your question. The expected molecular weight for the c-Kit precursor is ~ 120 kDa, and mature c-Kit is ~145 kDa. We have not tested this antibody, c-Kit (E-3): sc-365504, in mouse thymic epithelial cells so we do not have any information about the abundance of the protein in these samples. However, we would not expect c-Kit to be detected at a molecular weight ~55-60 kDa. We would strongly recommend including a secondary antibody control (omit primary antibody) to determine if the lower molecular weight band you are observing is non-specific due to your secondary antibody alone. And we would also recommend increasing the primary antibody concentration and using a dilution of 1:200. And using a positive control lysate for c-Kit, such as TF-1 or TT whole cell lysate, and loading 50 µg of protein.

Conditions of WB ; 8% SDS-PAGE, Wet transfer (400mA/2hr), 1:400 primary antibody in 5% TBST.
