BMP2 encodes bone morphogenetic protein 2 (BMP-2), a secreted ligand in the TGF-β superfamily that signals primarily through type I/II serine-threonine kinase receptors to activate SMAD1/5/8 and intersect with MAPK pathways. BMP-2 regulates osteogenic and chondrogenic differentiation, extracellular matrix remodeling, and developmental patterning, and it shapes cell fate decisions across mesenchymal lineages. Dysregulated BMP2 signaling has been associated with aberrant ossification, skeletal malformations, and altered tumor microenvironment cues that influence proliferation, migration, and epithelial–mesenchymal programs. BMP2 is therefore widely studied in bone biology, regenerative model systems, and pathway crosstalk analyses involving TGF-β/BMP signaling.
BMP-2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the BMP2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the BMP2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the BMP2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish BMP-2 protein expression.
This CRISPR knockout system enables efficient generation of BMP2-deficient cell models for investigation of BMP-2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
