SEE ALSO...

BLNK Antibody (2B11) is a mouse monoclonal IgG2a kappa light chain antibody that detects BLNK protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). BLNK (2B11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. BLNK protein, also known as B-cell linker protein, plays a crucial role in B cell receptor (BCR) signaling, which is essential for B cell activation, proliferation, and differentiation. Located primarily in the cytoplasm, BLNK acts as a central hub for signaling pathways initiated by BCR engagement, facilitating the recruitment and activation of downstream effectors such as GRB2, Vav, Nck, and phospholipase C gamma (PLC γ). BLNK′s location in the cell enables signal integration from various kinases, including Syk and Lyn, which are vital for proper B cell development and function. BLNK exists in two isoforms, pp68 and pp70, resulting from alternative splicing of the human BLNK gene transcript, and phosphorylation by Syk is a key regulatory step that enables the activation of downstream signaling cascades. BLNK serves as a significant target for studying B cell biology and related immunological processes.

How should the blocking peptide sc-8003 P be used?

Asked by: jenniferc
Thank you for your question. The blocking peptide is intended to be used as a negative control by preadsorbing BLNK (2B11): sc-8003 antibody with it prior to application. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization

Rated 4 out of 5 by from Potent and very specific antibodyThis monoclonal antibody was independently tested in-house by Kinexus Bioinformatics Corporation and validated for use for Western blotting. There was medium detection of the ~68+70 kDa target protein in mouse tissues (brain/testes/thymus/muscle mix), and no detection of the target in a HeLa/Jurkat cell lysate mix. No major cross-reactivities were observed at 2 µg/ml antibody concentration with off-target proteins in the mouse tissues tested or in the HeLa/Jurkat cell lysate mix. but a slightly high background was evident.
