Bim Antibody (H-5) is a mouse monoclonal IgG1 kappa light chain antibody, also known as BCL2L11 antibody, engineered to detect the Bim protein in mouse, rat, and human samples. Bim, or BCL2L11, is a crucial pro-apoptotic member of the Bcl-2 family located primarily in the cytosol and mitochondria, where Bim plays a vital role in regulating programmed cell death by antagonizing anti-apoptotic proteins such as Bcl-XL. Anti-Bim antibody (H-5) targets amino acids 4-195 of the human BimEL isoform, ensuring effective recognition of all three Bim isoforms, BimS, BimL, and BimEL, across multiple species. The localization of Bim is regulated, and under pro-apoptotic conditions, Bim translocates to the mitochondria, where Bim interacts with pro-survival Bcl-2 family members, influencing the intrinsic apoptotic pathway. Bim monoclonal antibody (H-5) is optimized for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA), making Bim (H-5) antibody an essential tool for researchers studying apoptosis and cell survival mechanisms. Understanding the cellular localization of Bim is important because Bim′s presence in the mitochondria is critical for the intrinsic apoptotic pathway, influencing key biological processes such as development, immune cell regulation, and cancer progression. Thank you for your question. We recommend the following amounts of protein for any western blotting experiment: 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane.

Are there any issues with background when using this antibody to stain mouse tissue or cells?

Thank you for your question. This antibody can be used to stain mouse tissue and cells. In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is currently available conjugated to HRP, PE, FITC, AF488 and AF647.

Can sc-374358 AC be used in Western Blot?

Thank you for your question. The agarose conjugated version of this Bim antibody, sc-374358 AC, is only recommended for IP or other applications where agarose is necessary. It is not appropriate for use in Western Blot.
