SEE ALSO...

β2-AR Antibody (E-3) is a mouse monoclonal IgG2b kappa light chain antibody that detects the beta 2-AR protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-β2-AR antibody (E-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The beta 2-adrenergic receptor (β2-AR) plays a crucial role in mediating the physiological effects of catecholamines, such as epinephrine and norepinephrine, which are vital for the regulation of cardiovascular function, smooth muscle relaxation, and metabolic processes. Located primarily in the plasma membrane of various cell types, including cardiac myocytes and bronchial smooth muscle cells, β2-AR is essential for the modulation of heart rate and airway dilation, making β2-AR a significant target for therapeutic interventions in conditions like asthma and heart failure. β2-AR′s structure, characterized by seven transmembrane helices, allows β2-AR to interact with G proteins, leading to the activation of intracellular signaling pathways that influence cellular responses. Understanding β2-AR signaling dynamics is critical for developing drugs that can selectively target this receptor to enhance therapeutic efficacy while minimizing side effects.

I am planning IHC study for β2- and β3-AR in adrenal medulla. WIth 1/100 dilution, how many immunostained slides could be made by 1 vial (200 µg/ml)?(The size of adrenal tissue section is about 1 cm X 1 cm.) Is the volume of product 1 ml?

I would like to know the quantity of product. I see the concentration (200ug/ml), but not the volume. Thanks.

Asked by: Timmy
Thank you for your question. Each vial contains 200 µg IgG2b in 1.0 ml of PBS with < 0.1% sodium azide and 0.1% gelatin. Can β2-AR (E-3): sc-271322 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Asked by: Cweed
Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, β2-AR (E-3): sc-271322 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 1 out of 5 by from Did not perform well for WBI obtained a sample of sc-271322 B2-AR (E-3) [Lot#K2216] to test on human myoblasts transfected with a non-silencing control siRNA or siRNAs specific to ADRB2, the gene that encodes the B2-AR. Immunodetection via western blotting did not produce a clear B2-AR-specific band. In contrast, detection of the same samples using sc-81577 B2-AR (R11E1) worked much more robustly.
