β2-AR Antibody (E-3) is a high quality monoclonal beta 2-AR antibody (also designated ADRB2 antibody, adrenoceptor beta 2 antibody, or B2AR antibody) suitable for the detection of the beta 2-AR protein of human origin. β2-AR Antibody (E-3) is available as both the non-conjugated anti-beta 2-AR antibody form, as well as multiple conjugated forms of anti-beta 2-AR antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Adrenergic receptors (ARs) (the term "adrenergic" reflects the alternative name for epinephrine, adrenaline) include four general types (a1, a2, b1 and b2) which are found in different target tissues and differ in their affinities and responses to various agonists and antagonists. cDNA clones have been isolated for all of the major AR subtypes and a number of closely related receptors have been identified by this approach. Each of the receptors have been shown to consist of single polypeptide chains which transverse the plasma membrane seven times, presumably forming a bundle of helices within the membrane. These transmembrane regions are hydrophobic and are interconnected by extracellular and intracellular hydrophilic loops. The coupling of ARs to specific intracellular effectors is mediated through diverse heterotrimeric G proteins and is regulated by G protein-coupled receptor kinases (GRKs), cAMP-dependent protein kinase A and protein kinase C directed phosphorylation. b2-adrenergic receptors bind cathecholamines (epinephrine, norepinephrine) and influence development, behavior, cardiac function, smooth muscle tone, and metabolism. b2-AR signaling complexes can contain C L-type calcium channel Ca(V)1.2, G protein, adenylyl cyclase, cAMP-dependent kinase, and PP2A phosphatase.

Can β2-AR (E-3): sc-271322 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, β2-AR (E-3): sc-271322 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Did not perform well for WBI obtained a sample of sc-271322 B2-AR (E-3) [Lot#K2216] to test on human myoblasts transfected with a non-silencing control siRNA or siRNAs specific to ADRB2, the gene that encodes the B2-AR. Immunodetection via western blotting did not produce a clear B2-AR-specific band. In contrast, detection of the same samples using sc-81577 B2-AR (R11E1) worked much more robustly.
