β-catenin Antibody (H-1) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 680-781 at the C-terminus of β-catenin, showing reactivity with human, mouse, rat, equine, canine, bovine, and porcine β-catenin in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) applications. β-catenin, also known as CTNNB1, is a multifunctional protein integral to cell adhesion and Wnt signaling pathways. β-catenin undergoes several post-translational modifications, including phosphorylation, ubiquitination, and acetylation, which regulate stability, localization, and activity. Phosphorylation of β-catenin by glycogen synthase kinase 3 (GSK-3) targets β-catenin for ubiquitin-mediated proteasomal degradation, a process critical for suppressing Wnt signaling in the absence of pathway activation. Conversely, when Wnt signaling is active, β-catenin accumulates in the cytoplasm, eventually translocating to the nucleus to regulate gene transcription. These modifications are essential for maintaining cellular homeostasis and controlling processes such as cell proliferation, differentiation, and migration. β-catenin also interacts with E-cadherin at adherens junctions, playing a structural role in maintaining tissue integrity. anti-β-catenin antibody (H-1) provides precise targeting of β-catenin′s C-terminal domain, making this reagent a valuable tool for studying post-translational regulation and functional roles in various cellular contexts.

Can β-catenin (H-1): sc-133240 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, β-catenin (H-1): sc-133240 has been validated for use in IHC with paraffin-embedded sections. Rated 5 out of 5 by from Suitable for endogenous beta cateninThis antibody works well, but in my case when I overexpress exogenous beta catenin with mutation on the S33, I am unable to detect the stronger band, even though the mRNA expression was evidently increased. I used the antibody at 1:500 dilution with MEF cells.
