β-Amyloid Antibody (B-4) is a mouse monoclonal IgG2a kappa light chain antibody that detects beta-Amyloid protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). β-Amyloid (B-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Beta-Amyloid protein, derived from proteolytic cleavage of Amyloid precursor protein (APP), plays a critical role in Alzheimer′s disease pathogenesis, as accumulation leads to neurotoxic plaque formation in the brain. These plaques are associated with neurodegeneration and cognitive decline, making beta-Amyloid research crucial for understanding and potentially treating Alzheimer′s disease and related disorders. Beta-Amyloid protein′s interaction with other proteins, such as APLP1 and APLP2, which are structurally similar to APP, highlights significance in cellular signaling and membrane dynamics. APLP2, a membrane-bound protein sharing homology with transmembrane-cytoplasmic domains of APP, further emphasizes interconnectedness of these proteins in neurodegenerative diseases.

I am a beginner in WB studies..so could you please let me know the difference between beta amyloid antibody D-11 and B-4 as I am confused for ordering an A-beta antibody.My samples are of mice and I need to study the expression of A-beta.

Thank you for your question. The "(D-11)" and "(B-4)" are alternate clone names for different antibodies that both detect the same β-Amyloid protein. Clone names are used to distinguish between antibodies and are used as unique identifiers. Both of these antibodies are suitable for detection of β-Amyloid protein in mouse samples. β-Amyloid Antibody (B-4): sc-28365 is recommended for detection of β-Amyloid and Amyloid A4 of mouse, rat, human and avian origin by WB, IP, IF, IHC(P) and ELISA. This is the more popular choice between these two antibodies, based on the number or product citations.

Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue? Or even double or triple staining on the same samples?

This antibody is currently available conjugated to phycoerythrin (sc-28365 PE), fluorescein (sc-28365 FITC), Alexa Fluor® 488 (sc-28365 AF488) or Alexa Fluor® 647 (sc-28365 AF647). The antibody had a great performance in every condition tested, using membrane visualisation with Super Signal West Pico PLUS Chemiluminescent substrate but also home reagents, on a Chemidoc Image Station.
