β-1,4-GalNAc-T2 Antibody (G-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects β-1,4-GalNAc-T2 in mouse, rat, and human samples by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-β-1,4-GalNAc-T2 antibody (G-1) is available in non-conjugated form and conjugated to agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® variants. β-1,4-N-acetyl-galactosaminyl transferase 2 (β-1,4-GalNAc-T2) consists of 566 amino acids and belongs to the glycosyltransferase 2 family, playing a crucial role in Sd(a) antigen synthesis, a carbohydrate determinant found on erythrocytes, colonic mucosa, and other tissues. β-1,4-GalNAc-T2 localizes to the Golgi apparatus membrane, where β-1,4-GalNAc-T2 transfers a beta-1,4-linked GalNAc to the galactose residue of an alpha-2,3-sialylated chain during Sd(a) production. β-1,4-GalNAc-T2 also completes the final step in Cad antigen biosynthesis, highlighting β-1,4-GalNAc-T2′s importance in cellular recognition and signaling. β-1,4-GalNAc-T2 expression is widespread, with highest levels in the colon and lower levels in kidney, stomach, ileum, and rectum. Mutations in β-1,4-GalNAc-T2 have been linked to Type I von Willebrand disease (VWD), the most common bleeding disorder in humans, characterized by reduced plasma von Willebrand factor levels. Two isoforms of β-1,4-GalNAc-T2 result from alternative splicing, demonstrating complex regulatory mechanisms in expression and function.
