Bcl10 Antibody (331.3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Bcl10 of mouse, rat, and human origin through applications such as western blotting, immunoprecipitation, immunofluorescence, immunohistochemistry, and enzyme-linked immunosorbent assay. Bcl10 (331.3) monoclonal antibody is offered in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. Bcl10, also known as CIPER, c-CARMEN, and mE10, plays a crucial role in immune signaling and was initially identified as a gene that is truncated or mutated in MALT B cell lymphomas and other tumor types. Bcl10 structure features an N-terminal caspase recruitment domain (CARD), which is essential for function in activating the NFκB signaling pathway via a NIK-dependent mechanism. This activation is significant as NFκB is a key regulator of immune responses and cell survival, making Bcl10 a vital component in the modulation of apoptosis and inflammation. Furthermore, Bcl10 has been implicated in the processing of caspase-9 to its active dimer, highlighting importance in apoptotic signaling. While Bcl10 is not mutated in many human tumors and lymphomas, overexpression can induce apoptosis through CARD-mediated oligomerization, underscoring potential as a therapeutic target in cancer research.

Does this vial contain only the light chain (and no heavy chain)? I am using this for immunoprecipitation and would like to know if I would see the ~50kDa heavy chain band on the western blot.

Thank you for your question. Bcl10 Antibody (331.3): sc-5273 is a mouse monoclonal primary antibody that contains the entire immunoglobulin molecule (heavy and light chains). If you use this antibody for immunoprecipitation of Bcl10, you would expect to see the heavy (~50 kDa) and light (~25 kDa) chains of the IgG if you are using a conventional anti-mouse IgG secondary antibody for detection in your Western blot. Because Bcl10 has an expected molecular weight of ~ 33 kDa, which is similar to the size of the IgG light chain, you may want to avoid using a secondary antibody in your Western blot. As an alternative, we would recommend using your primary antibody directly conjugated to HRP (sc-5273 HRP) if you are using a chemiluminescent detection system. The isotype of this primary antibody is IgG1, and the type of light chain is kappa. This information is provided on our website to help you select the appropriate secondary detection reagent, one of our recommended mouse IgG binding proteins, specific for kappa light chain.

I would like to use the Bcl10 Antibody (331.3) for IHC staining of mouse tissue. Is this possible?

Currently, Bcl10 Antibody (331.3): sc-5273 is available directly conjugated to HRP (sc-5273 HRP), phycoerythrin (sc-5273 PE), fluoroscein (sc-5273 FITC), Alexa Fluor® 488 (sc-5273 AF488) or Alexa Fluor® 647 (sc-5273 AF647).
