Bcl-2 Antibody (10C4) is a mouse monoclonal IgG1 kappa light chain antibody that detects Bcl-2 protein in mouse, rat, and human samples by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Anti-Bcl-2 antibody (10C4) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Bcl-2 plays a crucial role in regulating apoptosis, which is vital for normal development, maintaining tissue homeostasis, and providing defense against pathogens. This anti-apoptotic, membrane-associated oncoprotein promotes cell survival through interactions with other members of the Bcl-2 family, including both death suppressors like Bcl-xL and Mcl-1, as well as death agonists such as Bax and Bad. Bcl-2 contains a hydrophobic C-terminal domain that anchors to the mitochondrial membrane, effectively inhibiting cytochrome c release, a key factor in the apoptotic pathway. Post-translational modifications, including phosphorylation, can modulate Bcl-2′s anti-apoptotic activity. Mutations in the Bcl-2 gene have been implicated in various cancers, where normal apoptotic processes are disrupted, leading to uncontrolled cell proliferation.
